**********
Platinum Anti-Cancer Drugs
Platinum has the ability, in certain chemical forms, to inhibit the division of living cells. The discovery of this property in 1962 led to the development of platinum-based drugs to treat a wide range of cancers. Cisplatin, the first platinum anti-cancer drug, began to be used in treatment in 1977. Testicular cancer was found to be susceptible to treatment with cisplatin and there were other successes with ovarian, head and neck cancers.
Researchers at the Institute of Cancer Research and the Royal Marsden Hospital in London achieved a significant step when they found a compound similar to cisplatin in terms of activity, but much less toxic. This drug, carboplatin, was first approved in 1986. Recent research has sought to identify new platinum compounds which will treat tumours which do not respond to or which become resistant to cisplatin and carboplatin. The first of these drugs to reach commercialization is oxaliplatin, which is being marketed under the trade name Eloxatin.
Upcoming platinum anti-cancer drugs include satraplatin, which is being developed for treatment of prostate cancer. It is claimed that the use of satraplatin results in a higher survival rate than with existing chemotherapy treatments. Satraplatin will also be the first platinum anti-cancer drug that can be administered orally instead of intravenously, allowing patients to be treated at home. The drug is currently undergoing clinical trials.

• Platinum compounds continue to represent one of the major backbones
of modern chemotherapy in multiple cancers
(sales > $ 1b; typically used in combination therapy)
• Three platinum compounds on the market
- Cisplatin (i.v. infusion; off-patent in all major markets)
- Carboplatin (i.v. infusion; off-patent in most major markets)
- Oxaliplatin (i.v. infusion; expected peak sales: $ 1.5b)
• Satraplatin: very favorable toxicity profile vis-à-vis oxaliplatin & cisplatin;
only platinum analog with demonstrated activity in prostate cancer in a
randomized trial
• Upon market introduction of satraplatin: oxaliplatin only branded platinum
compound
- Satraplatin differentiated from oxaliplatin through oral activity,
benign toxicity profile and different tumor activity spectrum

August 19, 2002
SANOFI-SYNTHELABO has pulled a stroke ahead of troubled ImClone Systems Inc. in the race for the US colorectal cancer market. Last week the Food and Drug Administration (FDA) approved Sanofi`s Eloxatin (oxaliplatin for injection) after a 46-day priority review.
...
Lehman Brothers analyst Stuart Adkins projects peak sales of Eloxatin at $500 million, with additional indications. " Eloxatin could become the platinum compound of choice for use in combination therapy to treat all sorts of cancers, It will be very profitable for Sanofi and does not need a lot of marketing," says Mr. Adkins.

Sanofi-Synthelabo: more good news for Eloxatin
[Published: 26 January 2004 Source: Datamonitor Commentwire]
Sanofi-Synthelabo has submitted a sNDA for Eloxatin in the adjuvant treatment of colorectal cancer.
Sanofi`s [SAN.PA] supplemental New Drug Application for Eloxatin (oxaliplatin) in the US and Europe comes a week after Eloxatin was approved for the first line treatment of advanced colorectal cancer in the US. If approved, this new indication for the drug as an adjuvant treatment will give it a potential sales reach of $2 billion.
...
Eloxatin has been approved for second line treatment of colorectal cancer in Europe since 1996 and for first line since 1999 through a mutual recognition procedure. However, in the US the drug was only approved for second line treatment of advanced colorectal cancer in August 2002 and for first line in January 2004.
A decision is likely in 2005 and, if approved, the additional indication for oxaliplatin is likely to significantly increase the sales potential of the drug. Eloxatin is expected to achieve blockbuster status in 2004, but given the expansion in its indication, it has the potential to reach sales of $2 billion at its peak.

und was sagt GPC dazu?
